AmProtein CURRENT Perfusion Bioreactors
By using the CURRENT Bioreactor as a DO generator and suspension seed culture vessel, we have developed an extremely high-density cell culture system for protein and antibody production and alternatively, carrier-attached cells for vaccine production.

Features of the Perfusion Column
The perfusion column was pre-filled with affordable, single-use polymer fiber carriers. These perfusion column bags are shipped gamma-irradiated and ready-to-use once unpacked. The perfusion columns are affiliated attachments for the 5L, 50L and 150L CURRENT Bioreactors and are thus available in sizes according to bioreactor volume.

The most important features of the Perfusion Bioreactor include:
- Extremely high cell density in perfusion column filled with polymer fiber carriers reduces the need for serum and supports high production and high density during the high-speed perfusion.
- Harvest 1 bioreactor vessel volume (5L, 50L, 150L respectively) within 1 to 3 days, replacing a bioreactor 20 times larger in capacity. It is particularly suited for industrial applications.
- Besides its capabilities to produce large quantities of recombinant protein and antibodies, the perfusion bioreactor will replace current, outdated methods of vaccine production.
- Serum-free suspension CHO-K1 cells and attached CHO-K1 cells successfully tested for protein and antibody production.
- VERO, MDCK, Chicken, Fish cells, as well as several others have been successfully tested for animal and human vaccine production.

Conclusion: 150L paper-carrier perfusion bioreactor equals a 1500L fed-batch bioreactor.
Advantage: Single-use paper-carrier perfusion bioreactor is characterized by a high cell density, thus having strong cell growth and survival features. It is ideal for the industrial manufacture of protein and antibody drugs.

Conclusions:
1) A 50L paper-carrier perfusion bioreactor equals a production plant of 1200 large roller bottles and is perfect for larger-scale animal and human vaccine production, including chicken and fish. It is also great for rapid vaccine production of a deadly virus outbreak (such as SARS and bird flu outbreaks) after the virus DNA extracted.
2) Single-use and low cost.
Advantages:
1) Due to extremely high cell density, cell-cell growth support reaches the peak greatly for low-growth rate vaccine production cell lines;
2) Compared with roller-bottles and micro-carrier methods, seeding method (cell wash & trypsin-digest) for scale up is greatly simplified.
3) DO,pH,temperature control and cell line-specific medium optimization.
Mini Bioreactors to Support Process Development in Perfusion Bioreactors
AmProtein has developed a single-use, plastic miniature rolling bioreactor system for our customers to study the parameters involved in the process development stages for this perfusion system based on these polymer fiber paper carriers.
- Each minibioreactor vessel contains 20cm3 volume pre-filled with carriers and gamma-irradiated, ready-to-use.
- Each minibioreactor system can handle 45 vessels for efficient process development studies.
